- Melt agar in a microwave.
- Clean the lab bench with 70% ethanol.
- Using sterile technique, open the lid of the agar container.
- Transfer 30 ml of LB agar to a centrifuge tube.
- Add 25 mg/ml chloramphenicol and 50 mg/ml kanamycin.
- Cap the centrifuge tube and gently mix the solution by inverting the tube a couple times.
- Label petri dish.
- Pour the mixture into a petri dish.
- Partially cover the petri dish with lid to allow some moisture to escape.
- Repeat 5-9 until enough agar plates have been prepared.
